Adaçayı; Lamiaceae ailesine ait, aromatik, akdeniz kökenli bir bitkidir. Tüm dünyada, adaçayı, çeşitlihastalıklara deva bulmak ve hastalıklardan korunmak amacıyla yüzyıllardır geleneksel tedavidekullanılmaktadır. Adaçayı; sineol,borneol,rozmarinik asit, karnasol ve karnosik asitte dahil olmak üzerebir çok aktif bileşen ve esansiyal yağ içerir. Bu içeriği sayesinde antioksidan, antibakteriyel, anti-inflamatuar, antitümör gibi bir çok yararlı özelliğe sahip olmakla birlikte Alzheimer ve diyabet gibi çeşitlihastalıklarda da etkilidir. Bunun yanı sıra, Adaçayı bitkisinin bir çok kanser hücre hattı üzerinde etkiliolduğunu belirten birçok çalışma mevcuttur. Bu çalışmada ise adaçayı bitkisinin, birincil kemiktümörlerinin en malignant ve en yaygın görünen türü olan osteosarkoma hücreleri üzerindeki sitotoksiketkisinin incelenmesi amaçlanmıştır. Bu amaçla geleneksel tedaviden esinlenerek adaçayı infüzyonsolüsyonu olarak hazırlanmış ve U-2 OS osteosarkoma hücre hattı 3 gün boyunca çeşitli hacimoranlarında (1/1000, 1/100, 1/75, 1/50 ve 1/25 hacim/hacim) infüzyon solüsyonuna maruz bırakılmıştır.Bu süre sonunda hücre canlılığı nötral kırmızısı canlılık testi ile ölçülmüştür. Nötral kırmızısı canlılıktestine göre 1/1000, 1/100, 1/75, 1/50 and 1/25 hacim oranlarında verilen infüzyon solüsyonu U-2 OShücrelerinin canlılığını sırasıyla %3, %4, %26, %27 ve %29 oranında azaltmıştır. Elde edilen sonuçlar,adaçayı bitkisinin osteosarkoma hücreleri üzerinde sitotoksik etkisinin olduğunu desteklemektedir.
Sage is an aromatic, Mediterranean plant which belongs to the Lamiaceae family. All over the world,sage has been used in the traditional medicine for many centuries as a therapeutic and also apreventing agent for several diseases. Common sage possess many active components and essential oilsincluding cineole, borneol, rosemarinic acid, carnsole and carnosic acid. Due to these components andessential oils, Common sage has many properties such as antioxidation, antibacterial, anti-inflammation, anti-carcinogen and it is effective on various illnesses such as Alzheimer, diabetes etc.Furthermore, it was reported that common sage is effective for the killing of many type of cancer cells.In this study, we aimed to investigate the cytotoxic effect of common sage on Osteosarcoma which isthe most common and high grade malignant primary bone tumor. For this purpose inspired by thetraditional medicine common sage was prepeared as infusion solution and U-2 OS osteosarcoma cellswere exposed to infusion solution on various concentrations (1/1000, 1/100, 1/75, 1/50 and 1/25 (v/v)for 3 days. At the end of this time cell viability was measured by neutral red cell viability assay.According to neutral red assay the 1/1000, 1/100, 1/75, 1/50 and 1/25 (v/v) given infusion solutionswere reduced the viability of U-2 OS cells by nearly %3, %4, %26, %27 and %29 respectively. The resultsindicate that, sage has cytotoxic effect on osteosarcoma cells.
